INTENDED USE
The Parvovirus B19 IgM Enzyme Immunoassay is intended for the qualitative detection of IgM antibodies to Parvovirus B19 in human serum and plasma. The test is indicated for use in all women where there is suspicion of exposure to Parvovirus B19 to determine the presence of recent or current infection. In conjunction with the Biotrin Parvovirus B19 IgG Enzyme immunoassay this test maybe used as a component of clinical assessment of risk of foetal hydrops or foetal death.
ASSAY PRINCIPLE
The Biotrin International Parvovirus B19 IgM Enzyme Immunoassay is a mu-capture sandwich enzyme immunoassay for the detection of IgM class antibodies to Parvovirus B19 in human serum and plasma. Microassay wells are coated with a rabbit anti-human IgM. Total IgM antibodies in the serum or plasma will bind to the solid phase. Following a wash step, biotinylated purified Parvovirus B19 recombinant VP2 protein is added which binds to any human Parvovirus B19 IgM antibodies present. After another wash step, streptavidin peroxidase is added which binds to the biotinylated VP2 present. The whole complex is then detected by addition of substrate (TMB) which turns blue in the presence of peroxidase. A stable yellow end product is achieved by the addition of a stopping reagent.
